Vaccine
Background:
Many enveloped viruses, including influenza viruses(IFV-A & -B), respiratory syncytia virus (RSV), SARS-CoV-2 etc, pose great threat to human health globally. Smart enough, after their entry into host cells, the viruses hijack the host cells to fulfil their own life cycle. Remarkably, they express and anchor their viral envelop proteins on the host cell membrane. They then recruit components of the cellular ESCRT(endosomal sorting complex required for transport) system to mediate viral budding, forming new viruses with cell membrane-anchored envelop proteins (See below). The viral envelop proteins exhibit potent immunogenicity and are targets of many successful vaccines.
Our platform
Some special viral proteins alone can effectively recruit ESCRT for viral budding and form virus-like particles (VLP). Consequently, we create a matrix-derived VLP (mVLP) platform: we express a type of optimized viral matrix proteins for budding, together with viral envelope proteins of interest as antigen. The newly formed mVLPs retaining the immunogenicity of viral envelop proteins but non-parthogenic, become vaccine of ideal choice: potent and safe.
Following natural virus packaging path in vaccine-producing cells, co-expression of the optimized viral proteins for budding and viral envelop proteins as antigens leads to assembly, envelopment, budding and maturation, and eventually formation of virus-like particles (mVLP), devoid of viral genome.
